End-It™ DNA End-Repair Kit

End-It™ DNA End-Repair Kit用于将物理剪切或者限制性内切酶消化产生的5'- 和/或 3'-突出的双链DNA分子的突出末端补齐使之成为平端，使之能有效地克隆到质粒，粘粒，细菌人工染色体或其他载体中。这种转化是通过T4 DNA Polymerase的3'→5'核酸外切酶活性和5'→3'聚合酶活性实现的。试剂盒中还含有T4 Polynucleotide Kinase和ATP，可以使平端DNA的5'磷酸化以便于后续与克隆载体的连接。

End-It™ DNA End-Repair Kit可以进行20次末端修补反应（end-repair reactions）。采用Fast-Link DNA Ligation Kit末端补平的5'磷酸化的DNA可以有效地连接到平末端的去磷酸化的克隆载体中。

　

Applications Prepare sheared, nebulized, or restriction enzyme digested genomic DNA for blunt-end cloning into plasmid, cosmid, fosmid, or BAC vectors. Benefits The repaired DNA is blunt-ended and 5'-phosphorylated for immediate blunt-end ligation into plasmid, cosmid, fosmid, or BAC vectors. High specific activity T4 DNA Polymerase and T4 Polynucleotide Kinase are included for complete conversion of 5'- or 3'-overhang DNA to 5'-phosphorylated, blunt-ended DNA.

Figure 1. DNA fragments containing any type of ends are rapidly and efficiently converted to 5'-phosphorylated, blunt-ended DNA using the End-It™ DNA End-Repair Kit. The end-repaired DNA is ready for blunt-end ligation into the cloning vector of choice using, for example EPICENTRE's Fast-Link™ DNA Ligation Kit.

   RepliPHI™ Phi29 DNA Polymerase  

RepliPHI™ Phi29 DNA Polymerase (φ29 DNA Polymerase)是一种高效的有链取代活性的DNA聚合酶，此酶有3'→5'核酸外切酶活性，具有校读功能。

应用：

引物延伸。 

优点：

1， 有两种浓度可供选择1000U/ul和100U/ul。

2， 没有DNA污染。

3， 单独提供酶的稀释液。
